1. Examine the 3D Structure of a protein
a. Go to Protein Tools and select the PDBSEQRES:1gfl_A entry.
b. Click on the View Records button
c. The PDB Structure Explorer goes to the PDB web site





2. Protein Explorer
a. Click on the Protein Explorer
b. Scroll down, maximize the browser if necessary, click on the ¡¥Start Explorer Session¡¦ and then WAIT until the RED wait sign(lower right corner) becomes a GREEN ready sign



c. You can click the 'Hide/Show Water' button (Top left frame) to hide the red dots (water) in the right frame



d. Find the fluorophore
Enter the following commands in the text box in the lower left frame (press return after each entry):
Select ser65
Color yellow
Spacefill
Select tyr66
Color red
Spacefill
Select gly67
Color blue
Spacefill

Observations of some amino acid mutations: Ser65 to Thr increases intensity, Tyr66 to His shifts excitation to ultraviolet.
